The visualization of samples is a key tool in many fields of today's world. Scanning Laser Optical Tomography (SLOT) is a method to visualize mesoscopic bimomedical samples in 3D using fluorescence, scattering and extinction. In this thesis the applicability of SLOT has been pushed to new limits. First, SLOT was used to complement hearing restoration research and dental implant development. Furthermore, the SLOT setup is adjusted to meet the requirement of second harmonic generation (SHG). Now, noncentrosymmetric structures, as collagen, can be visualized with this intrinsic contrast mechanism inside the SLOT setup. Additionally, the SLOT setup was extended by new light sources and detectors. This way, techniques as spectral unmixing can be used for contrast enhancement. This makes SLOT the only mesoscopic volumetric imaging technique that provides in total four intrinsic contrast mechanisms (extinction, scattering, fluorescence and SHG).weiterlesen
